蚕砂光敏剂对肿瘤杀伤作用的超微结构研究

【摘要】 目的:了解蚕砂光敏剂对肿瘤组织的光敏效应。方法:通过蚕砂光敏剂作用小鼠移植瘤,给药量是小鼠体重50 mg/kg,尾静脉注射。给药后4小时用波长为660±10 nm,功率密度为≥200 mW/cm~2的红光照射20分钟,其后5分钟、30分钟、60分钟、2小时、4小时和24小时对小鼠移植瘤进行超微结构观察。结果:停止光照后5分钟,可明显观察到瘤区微血管裂解、瘤细胞固缩或解体死亡、细胞间充满变形的红细胞。结论:证明蚕砂光敏剂和血卟啉衍生物相似.对肿瘤组织具有杀伤作用,并对其机理进行了讨论,认为蚕砂光敏剂对肿瘤组织的杀伤机理,主要是对瘤区组织细胞的膜性系统的严重损伤。

【Abstract】 Objective: To investigate the photosensitive effect of a newly developed photosensitizer silkworm excrenemt (chlorophyll derivative CPD) on mice transplanted tumor. Methods: The mice were treated with a dosage of 50 mg/kg CPD(Vi). After 4h of injection, the transplanted-tumor mice received 20 min red light irradiation at wavelength of 660±10 nm and power density of ≥20 mW/cm~2. Then after respectively 5 min, 30 min, 60 min, 2 h, 4 h and 24 h, the mice were studied for their ultrastructure. Results: 5 min later, microvascular disintegration, cellular pyknosis or dissociation were obviously seen, and deformed erythrocytes filled the periphery of the tumor cells. Conclusion: It was shown that CPD and HPD were identical on injuring effect for tumor, and the mechanism of CPD was discussed with emphasis on its injure for membrane structure of tumor cells.
