老年细胞原癌基因c-fos、c-myc基因可诱导性减退与特异转录因子的关系
The decrease of induction of c fos/c myc expression by epidermal growth factor in relation to the specific transcription factor in human senescent fibroblasts.
【摘要】 目的观察人胚肺二倍体成纤维细胞的老年细胞中原癌基因c-fos、c-myc表达的变化及其与特异转录因子的关系。方法使用Northern印迹杂交法检测c-fos、c-myc基因表达情况,使用Southwestern印迹杂交法检测特异转录因子。结果表皮生长因子(EGF)对老年细胞中c-fos、c-myc转录的可诱导性下降,使与c-fos基因片段特异结合的蛋白质P91和与c-myc基因片段特异结合的蛋白质P80明显降低。结论老年细胞中c-fos、c-myc基因转录对EGF可诱导性下降,可能与其结合的转录因子活性改变有关。
【Abstract】 Objective To observe the changes of proto oncogene c fos/c myc expression and its relation to specific transcription factors in human senescent fibroblast after epidermal growth factor (EGF) addition. Methods The c fos/c myc expression were analysed by Northern blot. Transcription factors were analysed by Southwestern blot. Results (1)The expression of c fos/c myc was less susceptible to induction by EGF as the cells aged.(2)EGF increased the binding of protein P91 to c fos with regulatory sequence and P80 to c myc with regulatory sequence, but the increased levels of both proteins were significantly reduced in senescent cells. Conclusions Inability of c fos/c myc gene induction by EGF in senescent cells might be correlated with some DNA binding proteins.
