尼莫地平对新生猪缺氧缺血性脑损伤的防治作用

【摘要】 为探讨莫尼地平对新生儿缺氧缺血性脑病的防治作用,将86只新生猪分为正常对照组,缺氧缺血性脑损伤(HIE)1小时(1hr)、24hr、72hr组,尼莫地平0.5μgkg-1min-1治疗后1hr、24hr、72hr组,尼莫地平1μgkg-1min-1治疗后1hr、24hr、72hr组。检测各组红细胞胞浆总钙(RBCTCa)及游离钙(RBCCa2+i)的浓度变化。结果:HIE1hr组RBCTCa和Ca2+i均明显高于正常组对照(P<0.01),随着缺氧缺血状态的改善,RBCTCa和Ca2+i含量逐渐下降,HIE72hr组接近正常对照组(P>0.05)。两种剂量尼莫地平各治疗组RBCTCa和Ca2+i均明显低于HIE1hr和24hr组(P<0.01),与正常对照组比较,差异无显著意义(P>0.05)。提示:窒息后细胞内钙的沉积与新生猪HIE的发生有关,尼莫地平能阻止Ca2+内流,有效地保护神经细胞。

【Abstract】 The objective of this study was to investigate the changes in total calcium (TCa) and free calcium iron (Ca 2+ i) concentration in RBC cytoplasm in piglets and to study the pathogenesis of hypoxic ischemic encephalopathy (HIE) and protective effect of two different doses of nimodipine on HIE. TCa and Ca 2+ i of RBC were measured in 86 piglets which were divided into the following groups: normal control group ( n =9), post HIE groups [1 hour ( n =8), 24 hours ( n =8) and 72 hours ( n =8)], nimodipine (1 μg·kg -1 ·min -1 ) treatment groups [1 hour ( n =10), 24 hours ( n =10) and 72 hours ( n =9)] and nimodipine (0.5 μg·kg -1 ·min -1 ) treatment groups [1 hour ( n =8), 24 hours ( n =8) and 72 hours ( n =8)]. The results showed that RBC TCa and Ca 2+ i concentrations were significantly higher in HIE model 1 hour group than those in normal control group ( P <0.01) and then gradually reduced with time and approached to a level of the normal control group after 72 hours of HIE ( P >0.05). RBC TCa and Ca 2+ i were evidently lower in both different doses of nimodipine treatment groups than those in post HIE group ( P <0.01) and approached to a level of the normal control group ( P > 0.05 ). The results suggest that postasphyxiated intracellular calcium accumulation is associated with newborn HIE and that the calcium antagonist, nimodipine, is effective in protecting brain cells from further damage through its influence on cerebral calcium accumulation.
