高效液相色谱法测定大鼠血中头孢他啶浓度的实验

【摘要】 本文报告采用高效液相色谱法(HPLC)测定大鼠血中的头孢他啶浓度。样品在酸性条件下转入水相,样品中杂质在中性条件下转入混合溶媒。色谱柱为YWG-C18柱,内标物选用氢氯噻嗪。流动相为0.1mol·L-1醋酸铵缓冲液(pH4.59)与甲醇按88∶12配成的混合液。最低检出浓度为0.2μg·ml-1,日内相对标准偏差在3.3%以内,日间相对标准偏差在4%~12%之间。本法操作简便、快速、专一性强、重现性好。

【Abstract】 A HPLC method was established for the determination of ceftazidime(CAZ) in rat plasma. Ceftazidime was first extracted under acid condition into the aqueous phase and the impurity was extracted into the organic layer at pH 7. After addition of the internal standard(hydrochlorothiazide, IS),CAZ and IS was separated on a YWG C 18 column with a mobile phase of MeOH 0.1 mol·L -1 NH 4Ac(12∶88,v/v,pH 4.59 ).The minimum detectable concentration was 0.2 mg·ml -1 for plasma. The mean recovery of CAZ from plasma was 109%. The intra and interday coefficients of variation were< 3.3% and 4%~12%. This method is simple, rapid and accurate.
