糖基化低密度脂蛋白对细胞脂代谢及钙离子浓度的影响
The effects of glycosylated LDL on intracellular[Ca2+]and cholesterol deposition
【摘要】 观察糖基化低密度脂蛋白(glc-LDL)与人单核巨噬细胞相互作用对细胞脂代谢及钙离子浓度的影响。细胞降解 glc-LDL 较正常 LDL 组升高(P<0.05),结合 glc-LDL 具饱和趋势,亲和常数为17.82mg/L,最大亲和结合为434.78mg/g 细胞蛋白,细胞内总胆固醇含量升高(P<0.05),细胞游离钙离子浓度升高(P<0.05)。提示人单核巨噬细胞代谢 glc-LDL 途径与正常 LDL 不同,此过程伴脂质增加和钙离子浓度升高,可能为糖尿病病人易患动脉粥样硬化的原因之一。
【Abstract】 The changes of[Ca2+]and metabolism of lipid in human monocyte-macrophages as inter- acted with glycosylated LDL(glc-LDL)were observed.The intracellular[Ca2+]was higher than that of LDL(P<0.05).The binding internalization and degradation of glc-LDL were higher than that of LDL. The total cholesterol in the cell was higher than that of LDL(P<0.05).The results indicated that the process of metabolism of glc-LDL was different from that of LDL and accompanied by total cholesterol and [Ca2+]deposition.It made the cells easy to become foam cells which might be one of the causes that diabet- ic patients were feasible to atherosclerosis.
