高脂高胆固醇摄入对大鼠血脂、抗氧化能力及动脉内皮细胞的影响

【摘要】 用含猪油10%、蛋黄粉5%、胆固醇2%和胆酸钠0.5%的高脂高胆固醇饲料喂养健康雄性Wistar大鼠,观察此种膳食对大鼠生长、血清脂质含量、脂质过氧化物(LPO)、血浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、血栓素(TXB2)、前列环素(6-酮-PGF1α)、内皮素(ET)及肝脏和主动脉内皮细胞微观结构的影响,实验周期为12周。结果表明:(1)高脂高胆固醇摄入提高大鼠血清总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白胆固醇(LDL-C),降低高密度脂蛋白胆固醇(HDL-1);(2)高脂高胆固醇摄入使大鼠血清LPO明显升高,血浆SOD明显下降,而血浆GSH-Px虽有下降,但无统计学意义;(3)高脂高胆固醇组大鼠血浆TXB2及TXB2/6-酮-PGF1α(T/P)比值明显升高,而血浆6-酮-PGF1α明显下降;(4)高脂高胆固醇组大鼠血浆ET明显升高;(5)肝细胞病理检查亦显示高脂高胆固醇摄入可使肝脏脂肪蓄积严重,扫描电镜下可见高脂高胆固醇组大鼠主动脉内皮细胞损伤明显。提示高脂高胆固醇摄入可引起严重高脂血症及抗氧化能力下降和内皮细胞损伤等多种病变。

【Abstract】 To investigate the effects of high dietary fat and cholesterol on the serum lipids, antioxidative ability and aortic endothelial cell, 22 male Wistar rats Were assigned randomly to receive a normal diet (Control group, n = 11 )or a high fat and cholesterol diet (Study group, n = 11 ) for 12 weeks. The parameters included serum lipids, sezum lipoperoxides(LPO), plasma superoxide dismutase (SOD), glutathion peroxidase (GSH-Px ), thromboxane (TXB2 ), prostacyclin (6-keto-PGF1α ) and endothelin (ET). Histology of liver and aortic endothelial cell uilrastructure were also observed. As a result, The levels of serum total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C),serum LPO, plasma TXB2, TXB2/6-keto-PGF1α ratio (T/P), and plasma ET increased, and the high density lipoprotein cholesterol (HDL-C) level, plasma SOD activity, plasma 6-keto-PGF 1α level decreased, while plasma GSH-Px activities were not altered in study group when compared with control group. Severe fat accumulation in live and obvious damage of aortic endothelial cell under scan electron microscope were also observed in the study group. These results indicated that the high fat and cholesterol diet might cause hyperlipidemia, decrease the antioxidative ability and damage aortic endothelial cell.
