细胞毒T细胞对靶细胞的杀伤机制研究进展

【摘要】 <正> 细胞毒T细胞(cytotoxic T lymphocyte,CTL)怎样杀伤靶细胞?早期研究发现某些受CTL攻击的靶细胞质膜上可见类似于受补体攻击所致的"空洞"。所以,长期以来,人们一直认为CTL杀伤靶细胞的机制与补体类似。新近资料表明二者存在较大差异,例如CTL击杀的靶细胞核DNA迅速裂解,补体溶解的靶细胞则否,可以说穿孔机制只是CTL效应机制的一部分。近十多年来,CTL的杀伤机制方面有不少令人鼓舞的进展。一、CTL杀伤时相(phases of killing)可分为四个阶段:

【Abstract】 So far, little is known about the complete mechanism that cytotoxic T lymphocytes (CTL) kill their target cells. Howerer, the lethal armaments which CTL deliver the fatal blow are yielding to intense research. Pathologically, target cells attacked by CTL appeared necrosis and/or apoptosis. Recent studies have demonstrated that they kill target cells by multiple mechanisms delivering different mediators. Two major kinds of mediators are the pore-forming protein, perforin and the serine protease family of granzymes. Fas ligand (FasL) on the surface of CTL may be another molecular candidate which triggers cell death through binding to Fas antigen on target cells. It is believed that all the mediators acted synergistically. The following will briefly focus on: (ⅰ) the phases of killing during cellular cytotoxicity, (ⅱ) the biochemical and molecular features of perforin and serine granzymes, (ⅲ) the FasL pathway.
