血清中罗红霉素的反相高效液相色谱法测定

【摘要】 报道用高效液相色谱法测定血清中罗红霉素浓度.采用国产YWG—C18不锈钢分析柱(10μm,250mm×4.6mm),Waters486可变波长紫外检测器,以乙腈—甲醇—0.05mol/L磷酸(442036,用氨水调pH至7.2)为流动相,红霉素作内标.血清样品碱化后用二氯甲烷提取浓集,再经进一步纯化后进样,在210nm波长处检测,内标法定量.本法具有快速、简便、灵敏、准确等特点,适用于罗红霉素血药浓度测定及药代动力学研究.

【Abstract】 This paper reports a sensitive and rapid method for the determination of roxithromycin(RM)using erythromycin(EM)as the internal standard.RM and EM were extracted from alkalinized serum sample(500μL)with methylene chloride.After evaporation of the organic solvent,the residue was dissolved in 100μL of acetonitrile-ammonium phosphate and washed with n-hexane,then 50μL was injected onto a column(25cm×4 6mm)of YWG-C 18 (10μm).The flow rate of the mobile phase,acetonitrile-methanol-0 05mol/L phosphoric acid(44∶20∶36,adjusted to pH7 2 with ammonia water),was 1 2mL/min.The UV detector at 0 003 AUFS was set at 210nm.The retention times for RM and EM were 13 27min and 6 36min,respectively.Standard curve was linear in the concentration range of 0 25 to 16mg/L.The detection limit in serum was 0 06mg/L.The average extraction recovery was 68 8%;method recovery 102 8%;inter-day RSD less than 5 5%.
