阿霉素对培养大鼠内髓集合管细胞生长与增殖的影响

【摘要】 目的观察阿霉素对大鼠内髓集合管细胞生长增殖的影响。方法对大鼠内髓集合管细胞进行原代培养并给予阿霉素刺激,测定3H-TDR和3H-Leu的掺入。结果亚毒性剂量的阿霉素刺激细胞的生长,毒性剂量的阿霉素则抑制细胞的生长,且两种剂量的阿霉素均诱导细胞的肥大。结论在阿霉素肾病模型时的钠潴留及阿霉素诱导的肾小球硬化中,致病因子对小管细胞的直接作用可能发挥功能

【Abstract】 Objective By demonstrating the effect of adriamycin (ADR) on the proliferation and growth of rat inner medullary collecting duct (IMCD) cells in vitro to study the pathogenesis of ADR-induced nephropathy.Methods Primarily cultured rat IMCD cells were exposed to ADR of varied concentrations and incubated for different length of time.The cellular proliferation and growth were expressed by the number of cells and weight of cellular protein per flask and the uptake of 3H-TDR or 3H-Leu per well.Results Lower subtoxic-dose ADR stimulated,while higher toxic-dose ADR inhibited,the growth of IMCD cells in culture.Hypertrophy of cells was induced by both high and low doses of ADR.Conclusion It is likely that the direct action of ADR on renal tubule cells is involved in the development of ADR-induced nephrosis and ADR-induced glomeralosclerosis in rats.
