代用蛋壳培养体系对鸡胚骨组织中碱性磷酸酶活性的影响

【摘要】 以双黄鸡蛋的蛋壳为代用蛋壳,将正常孵化72h的鸡胚转移至其中进行孵化,取孵化至10、12、14、16、18和20d的鸡胚,测定其骨中碱性磷酸酶(ALP)的活性。同时分析正常孵化鸡胚骨中ALP的活性做为对照。结果表明:(1)对照组和实验组的ALP活性均随着胚胎日龄的增加而升高,尤其是16d后上升得很快;(2)14日龄以前的鸡胚,实验组与对照组相比,ALP活性无明显差异;(3)孵化后期同日龄鸡胚相比,实验组ALP活性高于对照组的ALP活性,差异显著。上述结果表明,在代用蛋壳中培养的鸡胚,发育后期钙的吸收不足而引起了骨代谢异常

【Abstract】 Recipient shells were prepared from double—yolked eggs We cut off the blunt end of the recipient egg shells and made a hole of 40mm in diameter Then chicken egg contents were discarded The fertilized eggs were normally incubated for 72h We transferred the chicken embryos into the recipient egg shells Chicken embryos in recipient egg shells were incubated until 10,12,14,16,18,20d,respectively We collected the leg bones(osfemoris) of embryos of different development stage and tested the activity of ALP(alkaline pholphatase)of bones From this experiment we can draw the following conclusions:(1)The activity of ALP of experiment group and control group tended to increase with the development of embryos After 16 days of incubation,it increased sharply;(2)During the late period of hatching the activity of ALP in experiment group was significantly higher than that in control group;(3)The activity of ALP of chicken embryos before 14 days between experiment group and the control significant difference was not found These conclusions indicated that the lack of calcium in embryos,incubated in recipient shells,leaded to bone metabolic abnormal
