基因免疫诱导9.1C3分子内影像类抗独特型抗体的产生
Induction of the Anti Idiotypic Antibody Bearing Internal Image of 9 1C3 Molecule by Gene Immunization
【摘要】 本文分别采用真核表达载体pEF-BOS及pCMV4构建含起始码和终止码的抗9.1C3分子单克隆抗体重链可变区基因重组表达质粒9.1C3VHpEF-BOS及9.1C3VHpCMV4,并将其分别免疫BALB/c小鼠。取免疫小鼠血清对K562细胞进行间接免疫染色和流式细胞仪分析表明,基因免疫小鼠体内可诱导9.1C3分子内影像类抗独特型抗体的产生。
【Abstract】 In this paper,the recombinant expression vectors 9 1C3 V H pEF BOS and 9 1C3 V H pCMV4 containing the complete open reading frame of 9 1C3 V H were constructed by using eukaryotic expression vectors pEF BOS and pCMV4 respectively BALB/c micc were immunized by using 9 1C3 V H pEF BOS or 9 1C3 V H pCMV4 The indirect fluorescent staining and flow cytometry analysis were performed, taking the serum from gene immunized mice as the first antibody and the K562 cells expressing 9 1C3 ligand molecules as the indicator cells The results show that anti idiotypic antibody bearing internal image type of 9 1C3 molecule can be detected in the serum from 9 1C3 V H pEF BOS or 9 1C3 V H pCMV4 vector immunized mice
