实验研究细胞因子在急性胰腺炎发病机理中的作用
Effect of Cytokines in Experimental Acute Pancreatitis
【摘要】 目的:观察大鼠血清中细胞因子在急性胰腺炎发病机理中的作用。方法:检测血清中细胞因子和TNF-α,观察胰、肝、肺、肾的病理变化及胰腺细胞培养液中淀粉酶和PLA2化。结果:在坏死性胰腺炎和水肿性胰腺炎大鼠血清中,IL-1β、IL-6、IL-2和TNF-α都明显高于正常对照组(P<0.01),水肿性和坏死性胰腺炎的胰组织中4种细胞因子都高于正常胰腺组(P<0.01),但两者之间未见明显差别。水肿性胰腺炎分离培养的胰腺细胞上清液中测出IL-6和TNF-α都高于正常胰腺组。结论:急性胰腺炎症过程中有多种细胞因子参与发病机理,其在体内放大作用造成胰腺本身在内的全身多器官功能损害,而且细胞因子的升高与疾病的严重程度相关。
【Abstract】 Background/Aims:To investigate of cytokines in the pathogenesis of experimental acute pancreatitis.Methods:SD rat acute pancreatitis models were examined:(1)serum IL-1β、IL-6、IL-12、TNF-α、mylase and PLA2;(2)morphology and pathology of the pancreas、liver、lung、and kidney;(3)cytokines in isolated pancreatic acini cells.Results:The mean serum IL-1β、IL-6、IL-12 and TNF-α levers in the acute edematous pancreatitis(AEP) group and acute nacrotic pancreatitis(ANP)group are significantly higher than those in control group(P<0.01),especially more higher in the ANP group.Similar results of cytokines in the pancreatitic tissue are seen.The release of TNF-αand IL-6 is increased in the inflammatory pancreatic acini cells.Conclusions:The cytokines may play an important role in the pathogenesis of ANP,and their release are also closely related to the severity of acute pancreatitis.
