CD3AK细胞联合IL-2脂质体抗鼠肝转移瘤的实验研究
Antitumor Effect of Combining Anti-CD3McAb Activated Killer Cells With Interleukin-2 Liposomes Against Murine B16 Melanoma Hepatic Metastasis
【摘要】 目的观察IL-2脂质体对CD3AK体内抗肿瘤的增强作用。方法:利用C57BL/6小鼠脾细胞与抗CD3单克隆抗体和白细胞介素———2(IL-2)共同培养制备CD3AK细胞(CD3McAbactivatedkilercels);从脾内注射B16黑色素瘤细胞制备鼠肝转移瘤模型:用PC和CH合成IL-2复合脂质体;CD3AK细胞联合IL-2脂质体用于体内抗B16黑色素瘤肝转移。结果:共同培养7天时CD3AK数即扩增到原来的8倍以上,明显高于LAK细胞(P<001),体外杀伤B16黑色素瘤细胞亦强于LAK细胞(P<0005)。CD3AK联合脂质体包裹的IL-2能显著减少肝转移瘤结节的数目(P<001),鼠外周血淋巴细胞明显增加(P<001);而同等剂量的CD3AK细胞加游离IL-2、游离IL-2及空白脂质体等组均无效。结论:本实验结果指示CD3AK细胞具有强的体内扩增能力及杀瘤活性,与IL-2脂质体联合具有体内抗肿瘤协同作用
【Abstract】 Aim:To observeted the antitumor efficacy of combining CD3AK cell with IL-2 Liposomes in a murine hepatic metastasis mode utilizing the B16 melanoma cells Method:The anti-CD3McAb activated killer cells(CD3AK cells) were induced by culturing C57BL/6 murine splenocytes with anti-CD3 monoclonal antibody and interleukin-2(IL-2).The B16 melanoma hepatic metastases models were established by injecting single tumor cells into spleen in C57BL/6 mice.Liposomes were prepared by using PC and CH.Result Expansion of cell numbers of CD3AK cells was 2.7 and 4.4 fold compared with LAK cells in 5 days and 7 days culture(p<0.001).Cytolytic activity of CD3AK cells was higher then LAK cells in vitro(P<0.005).The numbers of tumor hepatic metastasis were reduced significantly by combining CD3AK cells with IL-2 Liposomes in vivo(P<0.01) Conclusion:These results indicate that:(1)CD3AK cells are ease to be prepared and can generate large numbers effect cells,(2)the combination of CD3AK cells and IL-2 liposomes have synergistic antitumor efficacy in models of adoptive immunotherapy.
