卵巢癌细胞膜流动性与化疗增敏作用的相关性研究

【摘要】 研究化疗增敏剂三苯氧胺(TAM)、环孢菌素A(CsA)、水杨酸偶氮磺胺嘧啶(SS)、两性霉素B(AmB)、维拉帕米(VPM)及利尿酸钠(EA)对人卵巢癌细胞膜流动性的影响,探讨其与化疗增敏的关系。结果表明;①TAM降低肿瘤细胞膜流动性,AmB、CsA和SS增加膜流动性。随药物浓度增高,膜流动性改变增大。②TAM引起膜流动性降低与CDDP聚积量无关(P>0.05);而AmB、CsA和SS使膜流动性增加作用与之相关(P<0.05)。肿瘤细胞膜流动性改变可能是化疗增敏作用的一种机制,并希望成为寻找新的化疗增敏剂的又一途径。

【Abstract】 In this study, the effects of six chemosensitizers, tamoxifen (TAM), cyclosporin A (CsA),verapamil(VPM), sulphasalazine(SS), amphotericin B (AmB) and ethacrynic acid (EA) to the fluidity ofovarian cancer cell membrane were measured by fluorescent polarization. CDDP deposit in the cells wasdetermined at the same time. Results: ① TAM, AmB, CsA and SS all could sensitize CDDP, but VPM andEA couldn’t. ② TAM decreased the membrane fluidity, whereas AmB, CsA and SS increased it. The higherthe concentrations of the drugs were, the more the membrane fluidity was affected. ③ The decreasing of themembrane fluidity by TAM was not correlated with the amount of CDDP deposit in the cells; but the moreAmB, CsA and SS increased the membrane fluidity, the more CDDP collection in the cells there was; VPMand EA didn’t affect the membrane fluidity and there wasn’t change in CDDP collection in the cells.Conclusion: The change of the membrane fluidity of cancer cells by chemosensitizers could be a kind of mechanismof chemosensitivity, and this can be expected as another way to look for new chemosensitizers.
