慢性缺氧性肺动脉高压大鼠肺组织G_αmRNA表达的变化

【摘要】 为了探索慢性缺氧性肺动脉高压与G蛋白α亚基的关系,作者应用慢性低压缺氧方法(氧浓度10%,缺氧30~40d)建立肺动脉高压动物模型,再用核酸分子杂交方法测定大鼠肺组织Gα(G(αi-2)、G(αi-3)、G(αs))mRNA水平。结果表明,慢性缺氧性肺动脉高压大鼠肺组织GαmRNA水平与对照组比较,G(αi-2)、G(αi-3)分别为对照组的1.93和2.05倍;而Gαs无变化。提示,大鼠肺组织G(αi-2)、mRNA与G(αi-3)mRNA的增加,可能在慢性缺氧性肺动脉高压发病中起重要作用。

【Abstract】 To evaluate the relation between G proteins and hypoxic pulmonary hypertension (HPH), a stable rat model of HPH was established with chronic hypobaric hypoxia (10 % O2, 30-40d). By the use of dot-hybridization, the mRNA levels of α-subunits of G protein (Gαi-2, Gαi-3, Gαs) were examined in the lungs of rats before and after chronic hypoxia. The levels of Gαi-2,, Gαi-3, mRNA in HPH rats were 1. 93- and 2.05-fold respectively as high as those in normal rats, while there was no significant change in Gαs mRNA- The increased expression of Gαi gene might play an important role in hypoxic pulmonary hypertension.
