胺甲硫磷对大鼠血及脑乙酰胆碱酯酶的抑制作用

【摘要】 实验研究了胺甲硫磷在急性、亚慢性染毒时对大鼠血、脑乙酰胆碱酯酶(AchE)的影响,及在体外条件下,以人红细胞膜制备物为对象,脸甲硫磷抑制AchE的机理。结果表明,胺甲硫磷不仅在急性染毒时(1/10LD(50))对血、脑AchE有抑制作用,而且当亚慢性(90d)染毒时,高剂量组(在1/25LD50剂量下)对血、脑AchE也有抑制作用。体外试验表明,胺甲硫磷对人红细胞膜AchE的抑制作用具有剂量效应和时间效应关系,对AchE的抑制作用为不可逆性,其双分子速率常数较小,符合硫酮式有机磷化合物作用规律。

【Abstract】 Rats were exposed to organophosphorous pesticide o - methyl-o-(3-methyl - 4- methyl -thiophenyl ) thiophosphoramidiots at 0. 08 - 0. 16 mg/kg of dose by oral administration for 5 days (acute exposure) or 90 days (subehronic exposure)- It was found that acetylcholinesterase (AchE) activity of blood and brain decreased significantly both at 1/10 LD50 after acute exposure and at 1/25 LD50 after subchronic exposure- Furthermore, the effect of this pesticide on AchE activity of human erythrocyte membrane in vitro was studied. The result showed that this pesticide inhibited AchE rapidly and in a concentration-dependent fashion- The inhibition of AchE activities of human erythrocyte membrane was irreversible. The bimolecular rate constant (ki) is 107. 49 mol. min-1.
