【摘要】 采用戊二醛-明胶-乙二胺将氨基酰化酶交联固定在米曲霉细胞上,应用正交实验设计优选出相应的固定化条件,制备出具有较高反应活性和稳定性,适用于拆分DL-氨基酸的固定化细胞,并就pH、温度、离子浓度、底物浓度等因素对固定化细胞反应活性的影响进行了实验研究.分别在固定化细胞柱(ICC)和新型固定化细胞-膜耦联反应器(CI-CBMR)中进行了连续光学拆分N-乙酰-DL-丙氨酸的实验研究,结果表明该固定化细胞具有良好的工业应用前景.

【Abstract】 Aminoacylase was immobilized on the mycelium cells of Aspergillus oryzae by using ethylenediamine, gelatin and glutylalhyde. The proper immobilized condition was studied by orthogonal experimental design. The immobilized cells with excellent activity and stability for optically resoluting N acetyl DL alanine were obtained. The effects of pH, temperature, ion concentration and substrate concentration on the reactive activity of immobilized cells were studied. The continuous optical resolution of N acetyl DL alanine was investigated respectively in an immobilized cells column (ICC) and in a novel couple immobilized cells bed & membrane reactor(CICBMR). The results indicate that the immobilized cells are suitable for industrial applications.
