经S/D灭活病毒后新型凝血酶原复合物性质的研究
Study on Characteristics of A Newtype Prothrombin Complex Concentrate After the Virus Inactivation by S/D Method
【摘要】 对经有机溶剂/表面活性剂(S/D)处理后的新型凝血酶原复合物(PCC)与未经S/D处理的传统PCC进行对比分析,结果显示新型PCC不仅充分地保留了有效成分,且F-Ⅸ:C的比活性提高显著(P<0.01)。经SDS-PAGE、IE、CIE电泳及HPLC分析均反映出新型PCC的纯度有所改善,NAPTT和TGt50实验结果显示新型PCC明显优于传统PCC(P<0.05及P<0.01);采用兔Stasis模型进行Wessler试验,新型PCC的体内最小致血栓剂量>43PE/kg,其ED50至少高于传统PCC2倍以上。
【Abstract】 The characteristics of a newtype prothrombin complex concentrate (PCC) after being treated by S/D method were comparod with those of traditional PCC without treatment.The result showed that, not only the active components were maintained, but also the specific activity of F IX: C was significantly improved (p<0.01). All the analytic results by SDSPAGE, immuno-electrophoresis (IE), crossed immuno-electrophoresis (CIE) showed that the purity of the newtype PCC was significantly improved. The NAPTT and TGt50 of the newtype PCC were significantly longer than those of traditional PCC (P<0.05 and P<0.01respectively ). The potential effects of the 2 kinds of PCCs on thrombrosis were st udied in Stasis rabbit model and by Westem bolt. The result showed that the in vivo minimIll dose of the newtype PCC inducing thronbeis was>43PE/kg, and the ED50 of it was at least 2 timeshigher than that of the traditional PCC.
