硝基左旋精氨酸对睡眠抑制作用的机制研究
A STUDY OF THE MECHANISM OF L-NNA ON SLEEP INHIBITI0N
【摘要】 本文观察了硝基左旋精氨酸(L-NNA,50mg/kg,ip)和L-精氨酸(L-arg,110mg/kg,ip)对慢性植入电极的大鼠睡眠-觉醒周期的影响及中缝核5-羟色胺(5-HT)神经元免疫阳性反应的变化。结果表明:L-NNA显著抑制慢波睡眠和快眼动睡眠,使平均动脉压(MAP)升高。L-arg则使MAP显著降低,对睡眠无明显影响。预先给予L-arg可逆转L-NNA的效应。腹腔给予L-NNA后2h,中缝背核(DR)和中央上核(MnR)5-HT神经元免疫阳性细胞明显增多,与对照比较,P<0.01,此效应可被预先给予L-arg所减弱。提示L-arg:NO通路参与睡-醒周期节律的调节。中缝核头部5-HT神经元免疫阳性反应增强可能与L-NNA抑眠作用有关。
【Abstract】 In the present study, the sleep-waking cycle and the immunoreactivity of 5-HTcontainingneurons in rats receiving intraperitoneal injections of N-Nitro-L-arginine (L-NNA, 50 mg/kg) and L-arginine (L-arg, 110 mg/kg) were observed respectively.The results showed that injection of L-NNA suppressed both slow wave sleep (SWS) and rapid eye movement sleep (REMS) and increased mean arterial pressure (MAP). L-arg markedly attenuated MAP, whereas SWS and REMS were unchanged. The effects of L-NNA on sleep and MAP were reversed by preinjection of L-arg. In comparison with control group, the positive cells in dorsal raphe nucleus (DR) and raphe magUs nucleus (MnR ) increased 2 h a.fer injection of L-NNA. The effect was attenuated by preadministration of L-arg. All these results suggest that L-arg: NO pathway is involved in theregUlation of sleep-waking cycle and the increment of the positive immunoreactivity of the 5-HT-c0ntaining neurons in the DR may be related to the sleep-suppressive effects induced by L-NNA.
