【摘要】 目的研究白细胞介素２（ＩＬ－２）转基因乳腺癌细胞在小鼠体内成瘤性。方法用感染法将人ＩＬ－２基因体外导入乳腺癌细胞系４Ｔ１。转基因细胞４Ｔ１／ＩＬ－２培养上清内ＩＬ－２分泌量，ＤＮＡ、ＲＮＡ分别用ＥＬＩＳＡ，Ｓｏｕｔｈｅｒｎ杂交及逆转录ＰＣＲ法分析。体内实验将动物区分４Ｔ１，４Ｔ１／ＩＬ－２混合接种或单独接种三组。每周测量肿瘤大小，实验结束统计学用方差分析处理。结果４Ｔ１／ＩＬ－２细胞分泌的ｈＩＬ－２量为１２ｎｇ（每２４ｈ内１０６细胞），ｈＩＬ－２基因已整合到转导细胞基因组内并有ｈＩＬ－２ｍＲＮＡ表达。４Ｔ１／ＩＬ－２细胞接种动物后肿瘤发生延迟，瘤体小，与４Ｔ１接种组相比有显著性差异（Ｐ＜０．０５）。结论ＩＬ－２转基因细胞在动物体内引起抗肿瘤作用。更多还原

【Abstract】 Objective To study the tumorigenesis in mouse injected with hIL2 gene transduced murine breast carcinoma cell line.[WT5”HZ]Methods The hIL2 gene was transduced into murine breast carcinoma cell line 4T1 with the method of infection.The secretion of hIL2,DNA,RNA of IL2 transduced 4T1/IL2 cells were analyzed by ELISA, Southern blot and RTPCR.The tumorigenesis was studied in mouse injected with 4T1 mixed with 4T1/IL2,or 4T1,or 4T1/IL2 only.The size of tumors was measured weekly.Results One million of 4T1/IL2 cells could secrete up to 12 ng hIL2 in 24 hours.The hIL2 DNA had been inserted into the genome of 4T1/IL2 cells and had the expression of hIL2 mRNA.The tumorigenesis of 4T1/IL2 cells had been delayed and tumor size was reduced as compared with that of parental cells (4T1), (P<0.05).Conclusion IL2 gene transduced cells might activate certain local mechanism of tumor rejection in vivo.更多还原
