分泌抗TGEV单抗的杂交瘤细胞株在1.5L及新型5L细胞反应器中连续培养的研究
CONSECUTIVE CULTIVATION OF HYBRIDOMA CELLS WHICH COULD SECRETE McAb TO TGEV IN 1. 5L AND NEW TYPE 5L CELLIGEN
【摘要】 能稳定分泌抗TGEV单抗的杂交瘤细胞,培养于1.5L Celligen中,接种浓度不少于3×10~8/L,杂交瘤细胞在Celligen中生长,随着细胞浓度的变化,其生物学和动力学参数均发生相应的变化。杂交瘤细胞在pH7.18、DO50、50~70r/min、37℃及葡萄糖浓度为4.5g/L条件下生长良好,每天可收集1L单抗;又对改装后的5L Celligen的实验条件进行了初步探索,接种浓度为1.0×10~9/L、葡萄糖浓度维持在1.0g/L下每天生产单抗5L左右。
【Abstract】 Hybridoma cells which could stably secrete monoclonal antibody to TGEV were cultured in 1. 5L Celli-gen where the cell density was more than 3.0 × 108/L. The Kinetic and biological parameters also changed with the change of the cell density. The hybridoma cell grew well at pH7.18, DO50, 50-70r/min, 37℃ when the concentration of glucose in culture medium is 4.5g/L,about IL/day McAb could be collected. Furthermore, the culture condition of reequipped 5L Celligen was fundamentally researched vaccinated cell density was 1. 0 × 109/L, and 5L McAb could be collected every day under the control of stable concentration of glucose in culture medium with 1.0g/L.
