杀虫抗生素阿佛米丁发酵培养基筛选和产素代谢
OPTIMIZATION OF FERMENTATION MEDIUM FOR AVMPRODUCING STRAIN 86(X)-20(Streptomyces avermitilis) AND METABOLISM IN AVM PRODUCTION
【摘要】 AVM产生菌株86(X)-20(St.avermitilis)发酵培养基筛选结果表明:7种碳源材料中可溶性淀粉、淀粉和甘薯粉最佳,三者在菌丝生长量和AVM产量方面没有显著差别(P=0.05);7种氮源材料中,酵母膏、牛肉育、酵母粉最佳,三者之间没有显著差别。碳氮材料6种组合配方中,D1(甘薯粉4.0%十酵母粉0.6%)和D2(甘薯粉4.0%+酵母粉0.3%+蛋白胨0.3%)最佳。D1培养基50L小罐试验结果表明:接种后72h菌丝生长达到峰值时,AVM开始合成,合成最盛期为125~156h,发酵液正常pH值变幅为6.5~72.对还原糖和氨态氮代谢动态也进行了讨论。
【Abstract】 The test results of culture medium optimization for insecticidal antibioticavermectins-producing strain 86 (X)-20 (St’ avermitilis) ) indicated that soluble starch,starchand sweet potato meal exhibited the best effects on mycelian growth and AVM yields amongthe seven tested carbohydrate materials and there were no significant differences among thethree (P<0. 05); and so do the yeast ext., meat ext. and yeast powder among the seventested nitrogen source materials. D1 and D2 were the two best combinations of carbon andnitrogon sourcee materials among the six tested combinations. The results of experimentconducted in a 50 liters fermentation tank demonstrated:when the mycelian growth reachedthe maximum value 72 h after inoculation,the AVM can be detected and increased vigorously125 ~ 156 h after inoculation. The normal range of pH was from 6. 5 to 7. 2, the metabolicdynamics of reducing sugar and NH2-N were also observed and discussed.
