象面调制共焦扫描显微术应用研究
Investigation of the Application of Image Modulated Confocal Scanning Microscopy
【摘要】 对共焦扫描显微镜(CSM)的光瞳函数进行调制可以进一步提高分辨率,但实现却颇难.而象面调制和瞳面调制效果完全相同,并且容易实现.本文通过理论分析,得出了相干成象时象面调制和瞳面调制之间存在付里叶变换的关系.我们建立了由光电倍增管数据采集装置、步进电机驱动机械扫描装置和光学系统三部分组成的CSM实验装置.为一维相干照明条件下的象面调制CSM设计加工了状如Ronchi光栅的位相型膜片.实验结果表明,分辨率几乎提高了一倍,和理论分析吻合得很好.
【Abstract】 To enhance the resolution of a conventional confocal scanning microscope (CSM) by modulating its pupil functions is theoretically possible but difficult to realize in practice. It is known that image modulation has the same effect as pupil modulation and the former approach is more applicable. In this paper we demonstrate that for coherent case there is a relationship of Fourier transform between image modulation function and pupil function. We set up an experimental system for CSM made up of a PMT data acquisition unit, a steppingmotordriven scanning stage and the optics. A custommade optical phaseonly mask similar to the Ronchi grating is used for the 1D coherent image modulated CSM. The experimental results showed that the resolution is almost doubled twice and coincides with the theoretical prediction satisfactorily.
