同基因型葡萄愈伤组织脯氨酸累积变异系的抗盐性研究
Salte Tolerance of Proline Accumulation Variant Lines Come From Homogeneous Grape Somitic Callus
【摘要】 以同基因型葡萄的四个脯氨酸累积变异系为材料,研究了不同浓度NaCl胁迫下葡萄脯氨酸累积变异系的耐盐性。结果表明,葡萄脯氨酸累积变异系对高浓度NaCl胁迫有较强抗性。在240mmol·L-1胁迫下,对照系愈伤组织已停止生长,而4个变异系愈伤组织能较正常生长。在无盐胁迫下,变异系脯氨酸和可溶性蛋白含量均显著高于对照系,细胞膜透性显著低于对照系。在不同浓度NaCl胁迫下,4个变异系和对照系的脯氨酸含量、可溶性蛋白含量及细胞膜透性随NaCl浓度的增加而增加,但4个变异系增加幅度较缓,对照系增加幅度较大。在相同NaCl浓度下,4个变异系的脯氨酸含量、可溶性蛋白含量仍显著高于对照系,细胞膜透性显著低于对照系。4个变异系对NaCl胁迫的耐性较强
【Abstract】 Salt tolerance of four proline accumulation variant lines come from diploid grape callus was studied under different concentration of NaCl. The results showed that the NaCl tolerance of four proline accumulation variant lines was high. Under 240 mmol·L -1 NaCl stress, the control line callus didnt grew but four variant line callus could grow well. Under control conditions, contents of the free proline and the soluble proteins in four variant lines were significantly higher than those of control and the cell memberance permeability from the former was significantly lower than that of control. Under different concentration of NaCl,the free proline and the soluble protein contents as well as the cell membrance permeability in four variant lines and control line increased with the increase of NaCl concentration and increasing ranges of variant lines were lower than those of control. Under same concentration NaCl stress, contents of the free proline and the souble proten of four variant lines were significantly higher than those of control and the cell memberamce permeability of they were significantly lower than that of control.
