Percoll法处理对人精子运动参数的影响
Effect of mini-Percoll preparation on the movement parameters of human spermatozoa
【摘要】 为观察来源于3种不同精液标本精子运动参数的差异及其Percol处理后的变化,本文对新收集的正常精液精子(Ⅰ组),少精及弱精症精子(Ⅱ组)及正常精液冻融精子(Ⅲ组)各10份,分别于Percol处理前后用电子计算机精液分析仪(Computerasistedsemenanalyser)进行运动特性分析,记录MOT、VSL、VCL、LIN、ALH、及VAP6项参数,其结果进行统计学分析。Percol处理后,各组精子6项参数值均较处理前提高,差异具有统计学意义(P<0.05~0.001)。处理前Ⅰ组与Ⅱ、Ⅲ组除VSL无显著差异外,余5项参数值均有显著差异。Ⅲ组的ALH,VCL最高,其它参数Ⅰ组最高。Percol处理后差异值降低。Percol处理后精子运动6项参数值均提高;异常精液精子运动特性与正常精液精子有明显差异;正常精子冷冻复苏后其VCL及ALH明显提高。
【Abstract】 In order to observe the difference of the movement parameters of various human spermatozoa and the effect of Mini-Percoll treatment on them, the movement parameters of the spermatozoa from fresh normal semen(groupⅠ), oligozoospermia(groupⅡ) and frozen-thawing semen(Ⅲ) were analyzed with CASA before and after Percoll treatment respectively. The six parameters (MOT, VSL, VCL, LIN, ALH and VAP)were analyzed by statistical tests. The results showed that six parameters of three groups were promoted by Percoll treatment, the differences were statistically significant(P<0.05~P<0.005).Before Percoll treatment, there were statistical differences between the parameters of groupⅠ and groupⅡ, Ⅲ respectively except VSL. ALH and VCL were highest in group Ⅲ, and others (except LIN) were highest in group Ⅰ. These differences were still existed after Percoll preparation. The VCL and ALH of normal spermatozoa can be promoted by frozen-thawing.
