胎小脑移植对丙烯酰胺诱导大鼠小脑损害恢复的影响

【摘要】 腹腔注射丙烯酰胺(ACR)诱导SD鼠小脑损害,随机分组,分别给予小脑皮层内注射生理盐水、胎大脑悬液及胎小脑悬液,观察术后3个月内各组行为学、生化学及组织学的恢复:胎小脑移植组自主运动及共济失调的恢复明显优于其它两组;肌酸磷酸激酶(CPK)、谷胱甘肽转移酶(GST)活性的恢复在术后1、2、3月均高于其它两组,随时间延长差异越显著;小脑皮层组织学检查可见正常形态的蒲肯野细胞(PCs)较其它两组多见。结果显示,胎小脑移植对ACR中毒SD鼠小脑功能、生化及组织学的恢复有促进作用。

【Abstract】 Sixty six young adult female SD rats were administered acrylamide (30 mg/kg. body wt) intraperitoneally daily, five times a week for 3 consecutive weeks. Cerebellar functional deficiency was exhibited by a significant increase in gait scores, significant reduction in spontaneous locomotor activity, creatine kinase (CPK) activity and glutathione S transferase (GST) activity. 60 ACR treated SD rats were separated into three groups randomized. Group Ⅰ, cerebellar cortex received Ringer′s lactate solution injection merely. Group Ⅱ, neocortical suspensions were grafted into cerebellar cortex. Group Ⅲ, cerebellar suspensions were grafted. The recovery of neurobehavior, neurochemicals in three groups followed 1 month, 2 months and 3 months was studied. Group Ⅲ showed significant recovery in behavioral and neurochemical parameters. Recovery of these in group Ⅰ and group Ⅱ was relatively slow. The viability of cerebellar grafts was confirmed histopathologically. These results indicated that cerebellar transplantation could ameliorate ACR induced cerebellar function and biochemical deficits.
