γ 射线诱导人结肠癌细胞株p53和 WAF-1/CIP1 表达分析

【摘要】 WAF-1/CIP1是近年新发现的由p53激活的肿瘤抑制基因。本文以γ射线处理人结肠癌细胞株,分析DNA损伤诱导p53和WAF-1/CIP1在mRNA水平表达情况。结果表明,DNA损伤不能诱导mup53表达,也不能提高mup53细胞株WAF-1/CIP1的表达。但能强烈促进wtp53细胞株WAF-1/CIP1mRNA的表达,然而对wtp53表达的诱导在mRNA水平并不显著。这些结果提示:WAF-1/CIP1基因表达与wtp53产物有密切关系。本文对WAF-1/CIP1基因表达可能在判断肿瘤细胞p53是否突变以及选择最佳抗癌治疗方面的可能意义作了初步讨论。

【Abstract】 WAF 1 is characterized in recent years as a new tumor suppressor gene regulated by p 53 . This paper reports the expressions of p 53 and WAF 1/CIP 1 at mRNA level induced by γ irradiation in human colon cancer cell lines. Our results showed that DNA damage could not increase expressions of mutant(mu.) p 53 and WAF 1/CIP 1 in mu.p 53 bearing cell line. However,γ irradiation stimulated strongly WAF 1/CIP 1 mRNA expression, although wild type(wt) p 53 mRNA was not obviously elevated in wt p 53 bearing cell line. Thses results suggested that WAF 1/CIP 1 expession closely related to wt p 53 products rather than wt p 53 mRNA. The significance of WAF 1/CIP 1 expression was also discussed in the fields of judgement of p 53 mutation of tumor cells and selection of effective anti cancer therapy.
