维拉帕米对人单核巨噬细胞蛋白质、DNA、RNA合成的影响
Effect of Verapamil on Synthesis of DNA,RNA and Protein in Human MonocyteMacrophages
【摘要】 目的:观察维拉帕米对人单核巨噬细胞DNA、RNA及蛋白质合成过程的影响。方法:维拉帕米与人单核巨噬细胞孵育24h或4d,测定细胞3H-TdR、3H-UR、3H-Leu掺入人单核巨噬细胞状态及细胞内蛋白质含量。结果:维拉帕米在低浓度时对DNA、RNA合成有抑制作用,呈浓度依赖性;高浓度时作用明显,对RNA作用较DNA显著(P<0.05);对蛋白质合成在高浓度时有抑制作用。维拉帕米与成熟过程中人单核巨噬细胞作用4d,细胞蛋白含量显著降低(P<0.05),并呈浓度依赖性。结论:此作用可能为维拉帕米抑制细胞生长及动脉粥样斑块形成逆转心肌肥厚的机制之一。
【Abstract】 Objective:To observe the effects of verapamil on synthesis of DNA,RNA and protein.Method:When verapamil incubated with the cell for 24h or 4d,the incorperation of 3hTdR, 3HUR, 3HLeu into human monocytemacrophages was observed.Results:Verapamil inhibited the synthesis of DNA and RNA,showing a concentrationdependent effect.The effect on synthesis of RNA was more remarkable than that of DNA( P <0.05).The effect on the synthesis of protein was presented on the high concentration.When verapamil incubated with the cell maturing from monocytes to macrophages for 4d,the cells contained less protein than the compared group( P <0.05),and the effect was also of concentration dependent.Conclusion:Verapamil inhibited the growing and breeding of human monocytemacrophages,and it may be one of the causes that verapamil inhibits the formation of atheroma.
