黑曲霉HD9478纤维素酶固体发酵条件的研究

【摘要】 通过激光诱变筛选,获得一株高产纤维素酶的突变株HD9478,在以稻草、麦麸为主要原料的固体培养基上进行发酵培养,所产纤维素酶的固体曲,以羧甲基纤维钠为底物的纤维素CX酶活力为3500U/g,以滤纸为底物的纤维素CL酶活力为260U/g,以水杨素为底物的β-葡萄糖苷酶活力为1200U/g,发酵培养的条件是:培养基起始pH6.0~7.0,培养温度28~32℃,培养时间72h。三种酶作用的最适pH分别为3.5,5.5和4.5,最适温度分别为60,55,60℃,酶的pH稳定性与热稳定性为纤维素CL酶最高,纤维素CX酶次之,β-葡萄糖苷酶最差。

【Abstract】 A mutant strain HD9478 with high potency for cellulase production was obtained from industrial enzyme-producing strain which was screened by laser mutagenesis. In straw-wheat bran solid fermentation,its enzyme activity was 3 500 U/g on carboxyme thylcellulose-Na,260 U/g on filter paper and 1 200 U/g on salicoside. The optimum culture conditions were as follows:inifial pH6.0~7.0,temperature 28~32℃ and cutivation time 72 h. The optimal pH and temperature for cellulase reaction were pH3.5,5.5,4.5 and 60℃,55℃,60℃. The pH stability and thermal stability were studied. The results indicated that the stability of cellulase C L enzyme was the best and that of β-glucosidase was the worst.
