DIFFERENTIATION OF MAJOR GENOTYPES OF GIARDIA LAMBLIA(GINTESTINALIS) BY RESTRICTION ANALYSIS OF GENE AMPLIFICATION PRODUCTS
【摘要】 人们已经使用聚合酶链反应来扩增编码不同变异性表面蛋白的基因片断。9个瑞士蓝氏贾第虫无菌虫株被用于基因分析。根据产量、大小和限制性片断长度的多态性分析鉴定3个基因型。其中有5个蓝氏贾第虫株(01,B1,B2,B3-1A1和C1)被分类属于基因组Ⅰ。一个从人类获得的虫株(H2-17A)被分类属于基因组Ⅱ,它产生了一个特殊的VSP1267的PCR产物。这个虫株也产生了1.8kbtsp11和0.52kb类似tsa417/tsp11的PCR产物,这个产物具有基因组Ⅱ型虫株的限制性片断长度的多态性。3个虫株(O2-4A1,O3和H3-15K2)代表了一个新的基因型,它和基因组Ⅰ和基因组Ⅱ密切相关。这3个虫株显示了在tsp11PCR产物中完全相同的限制性片断长度的多态性,同时未能产生1个VSP1267PCR产物。这个结果表明能用这种方法划分肠道蓝氏贾第虫的不同基因型。
【Abstract】 The polymerase chain reaction (PCR) has been used to amplify segments of genes encoding different trophozoite variant surface proteins (VSPs).Nine Swiss isolates of Giardia lamblia were subjected to genetic analysis.Three genotypes were identified on the basis of product yield,size,and restriction fragment length polymorphisms (RFLPs).Five Giardia isolates (01,B1,B2,B3 1A1 and Cl from a sheep,three calves and a dog respecpively) were classified as belonging to genetic group Ⅰ.One human derived isolate (H2 17A) was typed as belonging to genetic group Ⅱ, yielded a vsp 1267 specific PCR product.This isolate also yielded 1.8 kb tsp 11 and 052 kb tsa 417/tsp 11 like PCR products possessing RFLPs typical of group Ⅱ organisms.Three isolates (02 4Al,03 and H3 15K 2 originating from two sheeps and a human respectively) represent a novel genotype that is closely related to genetic groups Ⅰ and Ⅱ.These three isolates exhibited identical RFLPs in their tsp11 PCR products and failed to yield a vsp 1267 PCR product.The results demostrate that different genotypes of G.intestinalis can be distinguished using this assay.
