小鼠胎肝基质细胞系MFLC的建立及鉴定

【摘要】 用体外长期培养的方法建立了小鼠胎肝基质细胞系(murinefetallivercel,MFLC),并运用免疫荧光技术、电镜技术、倍增时间的测定及染色体检查等方法,对此细胞系进行了细胞学鉴定。此细胞系建立至今已2年,历经3个阶段。细胞为多角形,用免疫荧光法显示胞浆内含角蛋白,同时也表达S-100和波形蛋白。电镜观察可见细胞形态多样,有些细胞含成束的张力丝,有些相邻细胞间有桥粒结构,部分细胞内可见病毒颗粒。细胞Ia表达呈强阳性,细胞倍增时间为24h。染色体为异倍体(50~78条),结果表明,本实验所建成的MFLC是以上皮细胞为主的已转化的胎肝混合细胞系。

【Abstract】 sing long term in vitro culture method, we established a murine fetal liver stromal cell line, called MFLC, which has been maintained for 2 years. The cell line was characterized by light microscope and electronic microscope′s observation immunofluorescent staining, doubling times′s determination and chromosome examination. The nature of MFLC was evidenced by the different shapes of the cells, Keratin expression of the cells, tonofilament in some cells′ cytoplasm and desomosomes between some cells. In addition, the cells were S 100 +, AV2 +, Thy 12 +, Ia +, Mac 1 -, DC -. Many virus particles wre observed in some cells′ cytoplasm, and all of the cells had abnormal number of murine chromosome. The doubling time of this cell line was 24 hours. These results provide evidence that MFLC cell line is a transformed murine fetal liver stromal cell line in which the epithelial cells are dominant.
