HeLa细胞G2/M/G1进程中PKA、PKC与CDC2活性变化相关性之初探
ELEMENTARY DETECTION OF CORRELATIONBETWEEN PKA, PKC AND CDC2 ACTIVITYIN G2/M/G1 PROGRESSING OF HeLa CELLS
【摘要】 为了研究上游信使系统中两个重要激酶——蛋白激酶C(proteinkinaseC,PKC)和蛋白激酶A(proteinkinaseA,PKA)与周期进程的相关性。采用胸腺嘧啶核苷(thymidine,TdR)双阻断和笑气(N2O)M期阻断相结合的方法,获得同步化的G2和M期细胞,并以此为模型,检测了信使系统中两个重要的蛋白激酶PKA、PKC以及M期引擎分子CDC2在HeLa细胞G2/M和M/G1过程中激酶活性变化情况,结果显示,PKC和PKA与CDC2活性变化分别呈正负相关性。
【Abstract】 おsing the method of thymidine (TdR) double arresting and N 2O arresting, synchronized G2 and M phase cells were obtained. Activity of PKA, PKC and CDC2 were assayed using this synchronized cell model. It indicated that PKA negatively while PKC positively correlate with CDC2 in G2/M/G1 progressing of HeLa cells.
