蛇毒神经生长因子的分离纯化及鉴定

【摘要】 采用SephadexG50、CM-Celulose32柱层析,从中华眼镜蛇中分离出神经生长因子(NerveGrowthFacter,NGF)。经SDS-聚丙烯酰胺凝胶电泳及Westernbloting法证明所得到的NGF为单一组分,相对分子质量约为1.3×104.经凝胶等电聚焦电泳测得NGF等电点PI约为7.0左右.经HPLC及电泳图像分析系统测得纯度为98%.此NGF经8d鸡胚背根神经节体外培养证明,具有促进神经纤维生长的活性.将所纯化的NGF免疫大白兔获得抗血清,经proteinA-Sephar-soeCL-4B亲和层析柱纯化获得了较为单一的抗NGFIgG

【Abstract】 Nerve Growth Factor (NGF) from the venom of Naja naja atra cantor was purified by Sephadex G50,CM Cellulose 32 and HPLC column chronatography.It was proved to be homogeneous by polyacrylamid lab gel electrophoresis,isoelectrofocusing electrophoresis, SDS polyacrylamid lab gel electrophoreis,and western biotting.Isoelectrofocusing point is about 7.0.It stimulates in vitro nerve fiber growth of dorsal root ganglia of 8 day chicken embryo.High titer antisera was obtained by injecting NGF in male rabbits.Homogeneous IgG was obtained from the antisera by protein A sephorose CL 4B affinity column chromatography.
