松茸的菌丝分离及纯培养研究
ISOLATION AND PURE CULTURE STUDIES ON Tricholoma matsutake
【摘要】 通过对松茸的组织分离、冷藏、培养条件进行研究,结果表明,子实体菌褶部位易于分离到松茸菌丝,子实体其它部位、菌根、菌窝土壤难于分离到菌丝。新鲜子实体是理想的分离材料,但低温保存60d的子实体,仍然可以分离到菌丝。松茸菌丝体在18~25℃下生长良好,22℃为最适生长温度。pH值以4.5~6.0较为适合,最适pH值为5.5。
【Abstract】 By studying the isolation and pure culture of T. matsutake , it shows that lamella is better for isolation than other parts of fruit body. And, it is difficult to obtain isolation from mycorrhiza and soil. Fresh fruit bodies are better than stored materials, but, after stored for 60 days, the fruit bodies can still be used for isolation. Studies on the culture condition of T. matsutake indicated that in pH 5.5 with temperature 22℃ gave optimum mycelial growth, whereas temperature among 28~32℃ showed lethal results in mycelial growth.
