分泌抗人IgMμ链McAb杂交瘤细胞株的建立
Establishment of Mouse Hybridoma Cells Secreting Anti-Human IgMμ Chain Monoclonal Antibody
【摘要】 本文从正常人(HBSAg阴性)血清中分离纯化人IgM,以此作为抗原免疫BALB/C小鼠,根据kohler和Milstein杂交瘤细胞建株原理,采用本室建立的常规方法,将免疫脾细胞和小鼠SP2/0骨髓瘤细胞进行融合,建立了分泌抗人IgMμ链单克隆抗体杂交瘤细胞株。经过间接ELISA双向琼脂扩散试验及封闭试验等方法证实,所分泌的抗体为抗人IgMμ链McAb,其中3A4、3D32株杂交瘤细胞经反复冻存、复苏及多次传代,均能分泌高效价抗体,且为IgG1亚类。本实验结果为抗人IgM鼠队嵌合抗体基因的构建奠定了基础。
【Abstract】 By fusing the sp2/0 myeloma cells with spleen cells from BALB/C mice immunized with human IgM, five strains of mouse hybridoma cells secreting anti-human IgM μ chain McAbs were successfully established. The specificity of McAb was identified by means of indirect ELISA. Of the five hybridoma cells, two strains (3A4、’3D3) secreting specific antibody against μ chain of human IgM were obtained by clonning for three times. After repeated frozen storage.thawed revival and generation they are still steadily producing high titer of McAbs. These McAbs were found to belong to subclass 1 of murine IgG.
