钝顶螺旋藻藻胆体的简易制备及褐藻酸钠对其稳定性的保护作用

【摘要】 以钝顶螺旋藻为材料 ,用改进的简易方法分离得到完整藻胆体 ,室温最大荧光发射峰位于 6 81 nm,加水解离后最大荧光发射峰蓝移到 6 5 5 nm,F6 80有两个峰 ,分别位于 6 38nm和 6 5 0 nm,从而表明藻胆体是完整的。用 436 nm波长激发 ,没有出现 Chla的特征荧光峰 ,F72 0的荧光激发光谱 ,也没有出现 Chla的峰 ,表明藻胆体溶液中不含 Chla。在藻胆体溶液中加入 1 %的褐藻酸钠 ,室温下保存 1 5 d后 ,光谱特性没有变化 ,藻胆体依然完整 ,表明褐藻酸钠对藻胆体的稳定性具有保护作用 ,可以延长完整藻胆体的保存时间 ,并对其可能的机理进行了讨论。

【Abstract】 The phycobilisomes in Spirulina platensis were isolated with a simple method, the maximum fluorescence emission of the phycobilisomes at room temperature was at 681 nm, 655 nm after dissociation in water, which indicated that the isolated phycobilisomes were intact. Moreover, there was no Chla in the phycobilisome solution. When 1 % sodium alginate was added into the phycobilisome solution and was stored for 15 days at room temperature, there were no changes in the fluorescence spectra, which suggested that the phycobilisomes still kept intact. The results showed that 1 % sodium alginate in the phycobilisome solution was helpful to prevent phycobilisome dissociation, and the mechanism was also discussed.
