【摘要】 报道一种快速、简单地从癌细胞中分离差异表达基因的改进的ｍＲＮＡ差异展示技术。选择两株具有相同的细胞来源，但具有不同转移能力的人肺腺癌细胞系ＡＧＺＹ８３ａ和Ａｎｉｐ９７３作为研究材料，利用一种灵敏度极高的ＳＹＢＲＴＭＧＲＥＥＮＩ荧光染料凝胶直接染色方法进行ｍＲＮＡ差异显示，对这两个细胞系的基因表达差异情况进行分析，发现在这两个细胞系之间存在明显的基因表达差异。对其中的两个差异片段进行了回收、克隆、序列分析及Ｎｏｒｔｈｅｒｎ印迹杂交，其结果提示这两种基因的表达或高表达与Ａｎｉｐ９７３细胞系的转移表型有关。本研究结果表明该方法具有快速、简便及重复性好等优点，适用于不同的对基因表达差异情况进行分析的研究。更多还原

【Abstract】 We have developed a rapid,simple and reliable method for isolation of differentially expressed genes from cancer cells.Two human lung adenocacinoma cell lines,AGZY 83a and Anip 973,were chosen as research materials,which originated from the same cells but manifested different metastatic potentiality.By the improved techniques of mRNA differential display and using of SYBR GREEN I,a high sensitive stain for detecting nucleic acid,we have studied the differences of gene expression between the two cell lines.The results showed that there were significant differences in gene expression between them.Two differentially expressed sequence tags have been cloned and analyzed.Our results indicated that the expression or over-expression of the two genes was associated with the metastatic phenotype of Anip 973 cells.更多还原
