新荧光衍生试剂N-羟基琥珀酰亚胺-α-萘乙酸酯的合成及其应用研究
Synthesis and properties of N hydroxy succinimidyl α naphthylacetate as a new fluorescence derivatization reagent for amino compounds
【摘要】 报道了新荧光衍生试剂N-羟基琥珀酰亚胺-α-萘乙酸酯(SINA)的合成。SINA的乙腈溶液在λex/λem=280nm/339nm处有最强荧光。在碱性水溶液中,SINA易与胺、氨基酸等氨基化合物进行衍生反应,生成有荧光的酰胺化合物,过量试剂在1h内完全水解为相应的α-萘乙酸,衍生产物及水解物易在反相键合色谱柱上分离。研究表明,SINA易制备,性能稳定,与氨基化合物易衍生,可用于柱前衍生HPLC分离-荧光检测氨基酸、生物胺等生物活性物质。
【Abstract】 A new fluorescence derivatization reagent for amino compounds, N hydroxysuccinimidyl α naphthylacetate(SINA),is easily prepared by the reaction of α naphthylacetic acid with N hydroxysuccinimide in the presence of dicyclohexylcarbodiimide SINA in acetonitrile presents strong fluorescence at λ ex /λ em =280nm/339nm The reagent can readily react with amino compounds to give the corresponding fluorescent amides, which can be separated on reverse phase column SINA is found to be a promising derivatization reagent for analysis of amino compounds by HPLC with fluorescence detection
