邻苯二胺与漆树漆酶反应的微量热法研究

【摘要】 在厌氧条件下,0.1mol/L的磷酸盐缓冲溶液(PH7.4)中,用微量热法测定了298.15K时漆树漆酶与邻苯二胺反应的摩尔反应焓△rHm=-293.58±0.29KJ/mol,米氏常数Km=0.103mol/L和表现一级反应速率常数k1=9.42×10-4S-1及此条件下邻苯二胺作底物时的漆酶活性值EA=O.1103IU.

【Abstract】 The total molar enthalpy and the Michaelis constant and the observed velocity constant of first-ordor reaction of the reaction between the Rhus ver-nicifera laccase and oPhenylenediamine have been determined at 298. 15K by microcalorimetry in 0. 1mol/L phos-phate salt buffer(pH7. 4)to be ΔrHm=-293. 56±0. 29KJ/mol and Km=0.103mol/L and K1=9. 42×10-4 S-1, respectively. The enzymatic activity of the laccase using the o-Phenylenediamine as the substrate have been determined to be EA=0. 1103(IU) in this condi-tion.
