L-丙氨酸酶法工业生产的研究

【摘要】 在800升气升式发酵罐上培养假单胞菌Pse.NX-1,获得高L-天冬氨酸-β-脱羧酶酶活,以L-天冬氨酸为底物游离细胞法转化生产L-丙氨酸,每升培养液可转化L-天冬氨酸2kg,最高达2.5kg,提取得到L-丙氨酸1.2kg,平均收率90%,产品质量达到美国药典XXⅢ版标准和日本味之素90版标准。

【Abstract】 Pseudomonas NX-1 was cultivated with 800 liter air-lift fermentor and high L-Aspartic acid-β-decarboxylase activity was obtained. For the enzymatic conversion of L-Alanine, the culture broth was employed as the enzyme source, and a large amount of aspartic acid (as much as 2000g/L, maximum 2500 g/L) was converted to L-Alanine. L-Alanine was easily isolated and 1200g dried product in 1 liter broth was obtained. The quality of L-Alanine conformed to the specifications provided in the American Pharmacopeia Rev. XX I or the Japanese Standard of Monosodium Glutamate Rev. 90.
