黑曲霉A3木聚糖酶固体发酵研究
Production of Xylanase by Aspergillus niger A3 Using Solid State Fermentation
【摘要】 筛选了一株高产木聚糖酶的黑曲霉(Aspergillus niger)A3菌株,研究了其在固体培养基中的发酵条件。该菌最适培养条件为:起始pH4.6,28℃,1ml孢子悬液接种量,蔗渣粉:麸皮为1.5:1,发酵3天。木聚糖酶活力可达5147IU/g培养基干重。氮源组成、温度、pH及发酵时间对曲中木聚糖酶和纤维素酶的比例有较大影响。与液体发酵相比,粗酶的最适反应温度均为55℃,最适反应pH值分别为4.6和4.2。在不同温度下保温1h,测得半失活温度(t1/2)分别为51℃和54℃。
【Abstract】 A high xylanase producing fungus Aspergillus niger strain A3 was isolated and its fermentation conditions by solid state fermentation were studied. The optimal culture conditions were as follows, initial pH 4.6, temperature 28℃, 1ml spore suspension inoculum, wheat brantbagasse = 1:1.5 and cultivations days. The xylanase activity was as high as 5147 IU/g dry medium. Nitrogen source Component, temperature, pH and cultivation time had great effects on the ratio of xylanse and cellulase. Some properties of the xylanase produced in liquid and solid cultures were compared. Both the optimal temperature were 55℃, optimal pH were 4 .6 and 4 .2 respectively. Xylanase synthetized in solid fermentation was little more themostatble than that from liquid cullture and the temperature for loss of half activity (t 1/2)in 1 h were 54℃ and 51℃ respectively.
