高效液相色谱法测定人尿液中氯氮平及其去甲基代谢物浓度

【摘要】 用高效液相色谱法同时测定尿液中氯氮平和其N-去甲基活性代谢物去甲氯氮平的浓度,测定方法采用ODS柱分离,紫外检测器检测波长为254nm,流动相为甲醇:水:四甲基乙二胺:冰醋酸(62:38:0.45:0.35,V/V),内标为安定。标准曲线范围为0~3000μg/L,(r=0.9999)。最低检出浓度:氯氮平4μg/L,去甲氯氮平2μg/L,尿液经二次溶剂提取后,氯氮平与去甲氯氮平的绝对回收率大于90%。日内变异与日间变异氯氮平为0.4~3.8%,去甲氯氮平为1.3~6.3%。本方法灵敏、精确,适用于尿液中氯氮平及去甲代谢物浓度的测定

【Abstract】 A HPLC method using ODS column and UV detector at 254 nm wavelength has been developed for determining clozapine(CLP) and its N-demethylated metabolite desmethylclozapine (DCLP) in urine. The mobile phase is consisted of methanol:water: tetramethy ethylemediamine:acetic acid(62:38:0.45:0.35 V/V). Diazepam is used as internal standard.Execellent liner relationship was obtained from the range of 0 to 3000 μg/L. The minimal limit of detection was 4 μg/L for CLP and 2 μg/L for DCLP. The recovery rate of CLP and DCLP were better than 90%;Interassay and intraassay evaluation was 0.4 to 3.8% of CLP and 1.3 to 6.3% of DCLP. Practice suggested this method was sensitive,accuracy and suitable for simultaneous determining CLP and DCLP in urine.
