激光微束法将外源基因导入大肠杆菌的初步研究
Primary Study on Introduction of the Tolerated Plasmid into E.coli Using Laser Microbeam
【摘要】 本文将Nd3+:YAG三倍频调Q激光微束系统用于质粒转化。以本实验室构建的耐药质粒作为导入基因,利用激光微束技术在大肠杆菌上打孔,通过高渗液的作用,在细胞内外形成渗透压压力差,将质粒成功地导入了大肠杆菌中,并获得了表达,取得了遗传性状稳定耐药菌株。通过导入、表达、筛选、检测等一系列工作,证明这是一种新的快速、高效的转化方法。
【Abstract】 :A laser microbeam system was introduced to the study of gene transfer.plasmid(pUH10)was used as a transrer gene,laser Laser microbeam was used to break up cell wall of E.coli,and hyperosmotic solution was used to form the osmotic pressure gradient,thus the tolerated plasmid was successfully introduced into E.coli and was experssed.The strain was genetically stable.This method is proved to be quick and efficent by introducing,expression,screening and detection.
