弓形虫体外感染巨噬细胞的实验研究

【摘要】 目的观察弓形虫体外感染巨噬细胞(macrophage,Mφ)及其在细胞内增殖的动态变化。方法用RH株刚地弓形虫感染小鼠腹腔Mφ,计算不同孵育时间的纳虫泡内虫体数,直线回归方程处理。结果弓形虫与Mφ的比例直接影响Mφ的感染率。弓形虫速殖子感染单层Mφ,只需1min便可侵入,虫体侵入细胞至开始增殖,约经3h的迟滞期,弓形虫增殖一代所需的时间为3.13h。结论RH株刚地弓形虫感染小鼠腹腔Mφ及其在细胞内增殖的动态变化,为进一步研究Mφ与弓形虫相互作用的机制提供了基础资料和体外实验模型。

【Abstract】 Objective\ To observe the dynamics of T.gondii infecting Mφ and its multiplication in Mφ in vitro.\ Methods\ The RH stain of T.gondii was used to infect murine peritoneal Mφ.\ Results\ The ratio of parasite to Mφ affected the infective rate of Mφ directly.It took only 1 min for parasite invading Mφ when contacting with the cells.Parasite in cells began its multiplication after a lagtime of about 3 h.\ By calculating the number of parasite in the parasitophorous vacuoles at different incubation times and linear regrssion,the mean time of a generation of T.gondii multiplicaton was 3.13 h.\ Conclusion\ This paper reported in detail the dynamics of T.gondii infecting murine Mφ and its multiplication in the cell,providing basic data and an experimental model in vitro for further investigation of the mechanism of interaction between Mφ and T.gondii.
