【摘要】 本文用高效液相色谱法(HPLC)研究了法莫替丁在肾功能减退犬的药代动力学及其在肾衰大鼠尿中的排泄。肾功能正常犬及其在肾功能减退时分别静注本品1.5mg/kg,结果表明,在肾功能减退时法莫替丁自犬体内消除减慢。正常肾功能大鼠及其在肾功能减退时分别静注本品0.8mg/kg 后,24小时由尿中排出的法莫替丁分别占给药剂量的64.9±4.5%及23.6±7.6%(p<0.001),二者差异显著。更多还原

【Abstract】 A reverse phase HPLC method for the determination of famotidine (Fa) in plasma and urine was developed.The method was simple,precise,highly specific and stable.The pharmacokinetic properties of famotidine in normal dogs and in dogs with renal insuffi- ciency was studied after administration.Following iv administration,plasma levels of Fa declined biexponentially and they were analyzed according to the two compartment open model consequently.The resullts demonstrated that the pharmacokinetics of Fa in dogs with renal insufficiency differed greatly from that in normal dogs.In dogs with renal in- sufficiency,the plasma elimination half life was markedly prolonged and the AUC was significantly increased,while the total body clearance was significantly decreased.In mormal rats unchanged Fa was predominately excreted into urine in normal rats.Fol- lowing iv 0.8mg/kgFa,the urinaty recovery of the unchanged form was 64.95±4.5%. But in rat with renal insufficiency,after same dosing,the mean culmulative renal excre- tion of Fa was calculated to be 23.6±7.6%.These results showed that renal excretion of Fa was significantly decreased in renal insufficiency.更多还原
