【摘要】 报道了利用聚合酶链式反应（ＰＣＲ）鉴定重组ＴＰＯ阳性克隆的方法，同时比较了该方法与酶切鉴定的结果．取转化克隆小量提取质粒琼脂糖电泳分析，对怀疑含有重组子的工程菌进行ＰＣＲ鉴定，结果阳性克隆得到了特异性扩增片段．该方法与酶切鉴定结果完全一致，其优点是简便、灵敏、特异更多还原

【Abstract】 The PCR method used to identify positive recombinant human thrombopoietin (TPO) cDNA clones is reported in this paper.It has also been compared with endonuclease identification.Electrophoresis of PCR amplified fragments in supernatant of lytic bacteria and fragments digested with endonucleases indicated that the target sequence can be specifically amplified in positive recombinant clones. This method is simpler,more specific and sensitive than the conventional method.更多还原
