【摘要】 以ＥＤＮ诱发大鼠实验性肝癌，同时连续１２周分别腹腔注射干扰素α－２ｂ（ＩＦＮ）、卡介苗（ＢＣＧ）、济南假单胞菌苗（ＰＴＶ）或ＩＦＮ和ＢＣＧ二者联用，对照组给予生理盐水。在诱癌过程第８、１２、１６周从同只大鼠体内分离出高纯度的腹腔巨噬细胞（ＰＭ）、血单核细胞（ＭＣ）、肺巨噬细胞（ＡＭ）和脾巨噬细胞（ＳＭ），并分别与人肝癌细胞联合培养，采用３Ｈ－ＴｄＲ掺入法测定免疫制剂对４种效应细胞抑人肝癌细胞增殖作用的影响。结果显示：①对照组诱癌大鼠４种效应细胞对人肝癌细胞增殖具有一定的自然抑制作用，ＰＭ作用最强，ＡＭ、ＳＭ、ＭＣ的作用依次减弱；②上述免疫制剂在体应用对４种效应细胞抑肝癌细胞增殖的增强作用，均以ＩＦＮ和ＢＣＧ二者联用的效果最佳，最强者可增强４倍以上；③单用免疫制剂者，ＢＣＧ的效果较ＩＦＮ强，ＰＴＶ的作用较弱。讨论分析了联用免疫制剂增强单核巨噬细胞抗肝癌最佳效果的机制及其实用意义。更多还原

【Abstract】 Hepatoma was induced in rats by administration of diethylnitrosamine(DEN).The rats were treated respectively by IFNα 2b,BCG,PJV,or combination of IFNα 2b and BCG,the controls were not treated with any immune stimulants(IS).The Monocytes(MC),peritoneal macrophages(PM),alveolar macrophages(AM),and spleen macrophages(SM) were isolated from the rats in the 8,12,16 weeks,and cocultured respectively with human SMMC 7721 hepatoma cells.Their cytostatic activity against human SMMC 7721 hepatoma cells were assayed through measuring 3 H TdR incorporation into tumor cell DNA.The results showed that ①The mononuclear macrophages of hepatoma rats (control group) possessed certain cytostatic effect against human hepatoma cells,the cytostasis of PM was the strongest ,the action of AM,SM,MC decreased successively.②The stimulative effects of combination of IFN α 2b and BCG on monounclear macrophages from four anatomic sites were more effective than those of single IS treatment in vivo.③The cytostatic effect of monounclear macrophages in experimental group was 5 times as high as that in the controls.BCG was more effective than IFNα 2b,the action of PJV is not as effective as that of other IS.The mechanism and significance of the combined use of IS for enhancing the cytostatic effect of mononuclear macrophages against hepatoma cells are briefly discussed.更多还原
