吗啡依赖大鼠脑细胞色素C氧化酶活性的改变
CYTOCHROME C OXIDASE ACTIVITY OF MORPHINE-DEPENDENT RAT BRAIN
【摘要】 目的:探讨吗啡依赖大鼠脑线粒体细胞色素C氧化酶(COX)活性的变化。方法:采用药参食法培养大鼠吗啡依赖模型,通过差速离心法分离线粒体。采用分光光度及化学发光等方法测定COX活性、心磷脂(CL)含量、膜流动性(MF)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:COX活性、CL含量、MF及SOD活性都明显降低(P<0.01),MDA含量明显升高(P<0.01)。结论:吗啡依赖大鼠脑线粒体COX活性降低,这与SOD活性降低导致的脂质过氧化水平升高有关。
【Abstract】 Objective: The present study probes the changes in cytochrome c oxidase (COX) in morphine-dependent rat brain. Methods: Morphine-dependence on rats was formed by drug-admixed foodrnethod. Brain mitochondria were separated by differential centrafugation. We measured COX activity,cardiolipin(CL) content, membrane fluidity (MF), superoxide dismutase(SOD) activity and malondialdehyde(MDA) content by the methods of spectrophotometry and chemiluminescence, and so on. Results: COX activity, CL content, MF and SOD activity all decrease significantly (P<0. 01);MDA content increase markedly (P<0. 01). Conclusion: The results suggest that COX activity has changed inmorphine-dependent rat brain, and the change may be related to the decrease in SOD activity leading tothe increase in lipid peroxidation.
