PCR-RFLP分析人结直肠癌DCC基因杂合性缺失
LOSS OF HETEROZYGOSITY INVOLV1NG THE DCC GENE IN HUMAN COLORECTAL CANCERS DEMONSTRATED BY THE POLYMERASE CHAIN REACTION
【摘要】 目的:探讨DCC基因在结直肠癌中杂合性缺失情况及其意义。方法:应用PCR技术检测了25例中国人散发型结直肠癌病人正常粘膜及癌组织和3个直肠癌细胞系DCC基因M2位点限制性片断长度多态性。结果:16例(64%)病人具有杂合性,其中4例(25%)癌组织表现出等位基因杂合性缺失。此4例均有淋巴结或肝转移。细胞系HCT,HR8348表现杂合性,HT29表现纯合性。结论:DCC基因是一个在结直肠肿瘤发生发展中起重要作用的抑癌基因,该基因的异常与结直肠癌发生发展有关。
【Abstract】 To investigate the significance of allele loss of DCC (deleted in colorectal cancer)gene in Chinese colorectal cancer patients. Methods: Using PCR technique, we examined 25 colorectalcancer patients and three cell lines for loss of heterozygosity (LOH) of DCC gene about M2 polymorphicsite. Results: sixteen of the cases (64% ) were informative, and among them 4 cases (25% ) showed allelic deletion. All the 4 cases had lymphatic or hepatic metastasis. One cell line showed homozygosity.Conclusion: DCC gene is an important tumor suppressor gene, and its alterations play a role in thepathogenesis and progression of colorectal cancer.
