感染新疆番茄的一种杆状病毒的鉴定

【摘要】 从新疆感病番茄分离到一株属于烟草花叶病毒组的病毒,感病番茄显示生长阻滞、叶片皱缩、花叶畸形以至坏死等系统浸染症状;该病毒可经汁液传播侵染属于10科的18种草本植物,但不能经桃蚜传播.感病组织粗汁液和提纯病毒制备物在SDS双向免疫扩散试验和酶联免疫吸附试验中与烟草花叶病毒UI株系抗血清发生强烈的免疫反应.该病毒的病毒粒子为长290nm,宽20nm的直杆状病毒粒子,其体外存活期在粗汁液中为2个月,在干组织中为3a以上,稀释眼点为106,致死温度为95℃.以聚乙二醇沉淀法和差速高心提纯了这病毒,提纯病毒(w=0.1%)的比消光系数E260=3.05,紫外吸收为典型的核蛋白吸收曲线,其A260/A280为1.25.这些结果证明该病毒是属于烟草花叶病毒组的烟草花叶病毒的一个株系,建议将此病毒株称为烟草花叶病毒新疆番茄株(Tobaccomosaicvirus-Xinjingtomato,TMV-XJTom).

【Abstract】 A tobamovirus was isolated from diseased tomato in Xinjiang. It was sap transmitted to 18 herbaceous species belonging to different botanical families and caused reduction in growth, leaf distortion, mosaic, malformation and necrosis in tomato. The virus was not transmitted by Myzus persicae. The sap from infered tissue and purified virus preparation strongly reacted with antiserum against tobacco mosaic virus U1 in SDS - agar gel double diffusion and enzyme linked immunosorbent assay. The modal virus particle was 290 um long and 20um wide. Longevity was more than 2 months in crude tobacco sap and more than 3 years in dry tobacco tissue, dilution end point was 106 and thermal inactivation point was 95℃.The virus was purified by polyethyleneglycol (PEG) precipitation and differential centrifugation.The extinction coefficient E260 was 3.05 for the virus isolate (w=0.1%). The purified virus had an ultraviolet absorption profile typical of a nucleoprotein, the A260/A280 ratio was 1.25. These results indicate that the virus isolate should be a strain of tobacco mosaic virus in tobamovirus group. The name of tobacco mosaic virus- Xinjiang tomato (TMV- XJTom) for this virus isolate was proposed.
